The air that one breathes may be contaminated by viruses that are airborne and may lead to the individual becoming ill. An example of an airborne disease is tuberculosis which is easily spread by air especially in crowded areas. Some individuals are allergic to specific foods, and if they take them by mistake, they become ill. Some diseases are waterborne, and if individuals take water that is contaminated they are a high risk of becoming ill. An example of a waterborne disease is cholera which greatly affects young children. Some diseases are caused by certain individual lifestyles such as eating habits. Eating foods that have high cholesterol levels and not engaging oneself in activities that help burn the cholesterol may lead to obesity and cardiac complications. Eating foods that have high sugar content may lead to high blood pressure and other related diseases. Some individuals have a lifestyle where they engage themselves in unprotected sex. Having unprotected sex is a very risky behavior as it makes one highly susceptible to contract sexually transmitted diseases and infections. Beyond Free Will in Shelly’s Frankenstein  Ã‚   One of the greatest gifts God has given to man is free will.   Free will is the ability to choose our own life’s path, to make decisions, and to suffer our own consequences. God has intended free will to allow us to live our own life by the rules we choose.   However, does free will reach a certain point as which to not crossover?   Man has always envied God, and has always tried to become god-like.   Does this ambition compromise our free will?   In Mary Shelly’s classic novel Frankenstein, Viktor Frankenstein’s tries to bring the dead back to life, and he is successful in animating a creation of his own.   The consequences of his ambition compromised his free will and destroyed his life.   Viktor Frankenstein reached the point of free will which man is not intended to cross over.   Viktor Frankenstein is a fool for trying to play God.   Ã‚  Ã‚  Ã‚  Ã‚  Ã‚  Ã‚   Free will was a gift granted to man right from the start of history.   In the story of Genesis, free will granted by God allowed Adam and Eve to eat from any tree in the garden, including the tree of knowledge.   However, God did set a rule.   â€Å"The Lord God commanded the man, â€Å"You are free to eat from any tree in the garden; but you must not eat from the tree of the knowledge of good and evil, for when you eat of it you will surely die.†1  Ã‚   Now, in this story, man is tricked by the serpent, representing the devil, and eats from the tree of knowledge.   He does not die in a sense, but is cast out of paradise and is forced to work off the land for the remainder of his now mortal life.   In a letter from Paul to the Galatians, Paul writes, â€Å"You, my brothers, were called to be free.   But do not use your freedom to indulge the sinful nature.†2  Ã‚   In thes... ...†9  Ã‚   Giving life is God’s job, and any man who tries to become god-like in this sense will surely suffer the consequences of his actions according to Mary Shelly.   I completely agree, and I will conclude with a retrospective quote from Viktor Frankenstein.   â€Å"Learn from me, if not by my precepts, at least by my example, how dangerous is the acquirement of knowledge, and how much happier that man is who believes his native town to be the world, than he who aspires to become greater than his nature will allow.†10 1. Narrative of Fredrick Douglass

The greatest thing about reading Narrative of the Life of Fredrick Douglass is that there are many different and interesting themes to learn from. Throughout Douglass’ story he teaches us many lessons and motifs, but one thing that stays constant is his belief in the fact that all men and women should be created equal, with equal rights without any constraints to his or her own individual freedom.The treatment of Douglass himself and the other slaves he worked with was unbearable and under such horrible circumstances that after reading his autobiography; it really makes me wonder what other types of things other slaves had to endure during their experiences. Narrative of the Life of Fredrick Douglass shows its readers that even though times can get extremely hard, there will still always be hope even when you think there is none. One of the most important ways slaves were kept in bondage was not simply the threat of physical brutality; rather, it was through deep and sustained ignorance.Slaves were not allowed to read and write and were therefore generally not aware of the events outside of the plantation, could not communicate with each other well to provoke rebellion or conduct escape plans, and could not reach the sense of self-sufficiency and pride that came from being educated. Literacy brought with it an understanding of the larger world. It opened up before a slave the idea of justice and an understanding of history. Reading the Bible led to a truer comprehension of Christianity. Douglass was able to first engage with abolitionism when he attained literacy.He also became fully aware of the reality of slavery; he wrote â€Å"[Literacy] had given me a view of my wretched condition, without the remedy. It opened my eyes to the horrible pit, but to no ladder upon which to get out. In moments of agony, I envied my fellow-slaves for their stupidity† (Page 56). Ignorance was thus a way for slaveholders to keep their slaves manageable, happy, calm, and content. Once a slave moved beyond such darkness into a world filled with understanding, he was only able to do what Douglass eventually did – attempt to escape from his ties.While reading Narrative of the Life of Fredrick Douglass it is almost like reading about how white men dehumanized their slaves. The first example of this is shown in Chapter 1 when Douglass’ mother passes away. â€Å"Never having enjoyed, to any considerable extent, her soothing presence, her tender and watchful care, I received the tidings of [my mother’s] death with much the same emotions I should have probably felt at the death of a stranger† (Page 43).This quote explains that Douglass, like many other slaves, never had much of any relationship with their birth mothers. Considering that some slaves are taken away from their birth mothers only a few short years after they are born it is easy to understand why Douglass felt this way. It’s extremely sad to read how a chi ld can feel almost no emotion after hearing of such a tragic loss. I imagine if that were to happen to me and how I would feel and can only feel disheartened by the fact that most slaves never got to have relationships with their mothers.Another example of such degrading behavior by the slaveholders is simply how they scarcely feed their slaves. They expect so much work and cooperation from them and think that any amount of food, big or small, will help them to get their work done. Slaveholders instill in the minds of slaves that being unfed, whipped and called awful names is the best kind of life they will ever have. These were horrible characteristics of slavery but were well depicted by Douglass in his autobiography. According to Nathaniel P.Rogers’ Southern Slavery and Northern Religion: February 11, 1844 (Page 139), it is explained that Douglass arrives to give a speech and to tell his story to an audience who is very apprehensive and uncertain about hearing him speak. H owever, they were very curious to see him. After giving his speech, which was well received by the audience, he was asked more and more questions and they wanted to know more about his life and journey through slavery in general. â€Å"There was great oratory in his speech-but more dignity and earnestness than what we call eloquence.He was not up there as a speaker-performing. He was an insurgent slave taking hold on the right of speech, and charging on his tyrants and bondage of his race† (Page 141). Reading this review makes me really happy to know that while he had an awful time as a slave, escaping and gaining freedom was the best thing that ever happened to him. It is satisfying to read that others can feel for Douglass, though not come close to imagining what he had gone through, but having some sort of sympathy and realization that he and many other slaves are great human beings with inspiring stories to share about he freedom they all deserve. He teaches us that while we all go through horrible things; there is always a silver lining and something to be learned. Narrative of the Life of Fredrick Douglass has really taught me a lot about myself and about a whole other kind of people that I really had no knowledge of. I truly believe that Douglass makes his audiences believe in what stands for; all men and women should be created equal, with equal rights without any constraints to his or her own individual freedom. This autobiography has a lot to teach a person and I would recommend it to anyone.It is thoroughly enjoyable and is the kind of book that makes you look past just the text. It makes you think about your own life, putting it in perspective and realizing what is really important. It is an interesting story that not many have heard themselves and really know little about its topic. I suggest to anyone who wants to read this narrative that they keep an open mind no matter what their previous views, religious or not, are so that they can ful ly understand and accept this person’s journey through slavery.

Louisiana Serial Killer Ronald Dominique

Louisiana Serial Killer Ronald Dominique Ronald J. Dominique of Houma, LA has confessed to murdering 23 men over the course of nine years and dumping their bodies in sugarcane fields, ditches and small bayous in six southeast Louisiana parishes. His reason for killing? He did not want to return to jail after raping the men. The First Victims In 1997, authorities found 19-year-old David Levron Mitchells murdered body near Hahnville. The body of 20-year-old Gary Pierre was found in St. Charles Parish six months later. In July 1998, the body of 38-year-old Larry Ranson was found in St. Charles Parish. Over the next nine years, more bodies of men ranging in age from 19 to 40 would be found dumped in sugarcane fields, desolate bayous, and ditches in remote areas. Similarities in 23 of the murders lead investigators to suspect the men were victims of a serial killer. The Task Force A task force made up of nine South Louisiana parish sheriffs offices, the Louisiana State Police and the FBI were formed in March 2005, to investigate the murders. Investigators knew the 23 victims were mostly homeless men, many who led high-risk lifestyles, which included drug use and prostitution. The victims had been asphyxiated or strangled, some raped and several were barefooted. The Arrest After receiving a tip, authorities armed with forensic evidence, arrested Ronald Dominique, 42, and charged him with the murder and rape of 19-year-old Manuel Reed and 27-year-old Oliver Lebanks. Just days before his arrest, Dominique had moved from his sisters home into the Bunkhouse shelter in Houma, LA. Residents of the home described Dominique as odd, but no one suspected he was a killer.​ Dominique Confesses to 23 Murders Soon after his arrest, Dominique confessed to murdering 23 southeast Louisiana men. His tactics in capturing, sometimes raping then murdering the men was simple. He would lure homeless men with the promise of sex in exchange for money. Sometimes he would tell the men he wanted to pay them to have sex with his wife and then show a picture of an attractive woman. Dominique was not married. Dominique then led the men to his home, asked to tie them up, then raped and eventually murdered the men to avoid arrest. In his statement to the police, Dominique said the men who refused to be tied up would leave his home unharmed. Such was the case with one unnamed man who reported the incident to the task force, a tip that eventually led to Dominiques arrest. Who Is Ronald Dominique? Ronald Dominique spent much of his youth in the small bayou community of Thibodaux, LA. Thibodaux sits between New Orleans and Baton Rouge and is the type of community where everyone knows a little about each other. He attended Thibodaux High School where he was in the glee club and sang in the chorus. Classmates who remember Dominique say he was ridiculed for being homosexual during his teen years, but at the time he never admitted he was gay. As he got older, he seemed to live in two worlds. There was the Dominique who was helpful to his neighbors in the small trailer parks where he lived. Then there was the Dominique who cross-dressed and did bad impersonations of Patti LaBelle at the local gay club. Neither world embraced  him, and among the gay community, many remember him as someone who was not particularly well liked. Through most of his adulthood, Dominique struggled financially and would end up living with his mother or other relatives. In the weeks before his arrest, he was living with his sister in a single-wide trailer. He was suffering from declining health, having been hospitalized for a severe heart condition and forced to use a cane to walk. Outwardly, there was side to Dominique who enjoyed helping people. He joined the Lions Club just months before his arrest and spent Sunday afternoons calling out Bingo numbers to senior citizens. The membership director said he was well liked by everyone he had met through the Lions Club. Maybe Dominique had finally found a place he felt accepted. What sparked Dominique to move from the comfort of his sisters home to the dismal surroundings of a shelter for the homeless is uncertain. Some suspect the family grew uncomfortable by the 24-hour police surveillance and Dominique, knowing he was soon to be caught, moved away to avoid getting his family involved in his arrest. A Criminal History Dominiques past arrests include forcible rape, disturbing the peace and telephone harassment. Feb. 10, 2002 - Arrested in Terrebonne Parish after he allegedly slapped a woman during a Mardi Gras parade. According to the reports, Dominique accused a woman of hitting a baby stroller in a parking lot. The woman apologized, but Dominique continued to verbally assault her, and then slapped her across the face. He was arrested but entered a parish offenders program instead of standing trial. Reports show he met all his conditions in the program in October 2002.May 19, 2000 - He received a summons to appear in court on disturbing the peace charges. Since it was a misdemeanor, he was able to plead guilty and pay a fine to avoid appearing in court.Aug. 25, 1996 - Dominique was arrested on forcible rape charges and booked on a $100,000 bond. According to neighbors, a partially dressed young man escaped from the window of Dominiques home in Thibodaux, screaming that he had tried to kill him. When the case was brought to court, the victim could not be found to testify. In November 1996, the judge continued the case indefinitely. May 15, 1994 - Arrested and charged with driving while intoxicated and speeding.June 12, 1985 - Arrested and charged with telephone harassment. He pleaded guilty, paid a $74 fine and court costs. Three days after Dominiques arrest for killing Mitchell and Pierre, investigators said Dominique confessed to 21 other murders, giving details only the killer would know.

However, despite years of desegregation, social and cultural differences still exist. One of these differences that still exists is in the institution of marriage. Americans have been and are continually moving slowly away from segregation. In the past forty years, a multitude of changes have transformed schools, jobs, voting booths, neighborhoods, hotels, restaurants and even the wedding altar, facilitating tolerance for racial diversity (Norman 108). In the 1960's, when housing discrimination was outlawed, many African-Americans moved into mainly Caucasian neighborhoods. The steadily growing areas in the west and southwest are least segregated, because these areas never had the entrenched African-American and Caucasian sections of town (Up For Separatist 30). Even more visible signs of desegregation can be seen in the areas of education. A study done by the University of Michigan shows that integration on campuses occurs on a regular basis. The racial lines are crossed routinely; about 50% of African-Americans and 15% of Caucasians reportedly study together. Eating patterns also share the same similarities. At a social level, there has been a steady convergence of opinion on a variety of racial issues. Since 1972, surveys have asked whether the respondent would favor a law making inter-racial marriages illegal. In 1980, the results showed that 30.1% of Caucasians and 18.3% of African-Americans favor such a law. By 1994, the collected data showed 14.7% and 3.2% respectively. Married to Maryann Burke at nineteen, and having two children in the space of seventeen months, the Carvers life was decided for years to come. We thought we could bow our necks, work very hard, and do all that we set our hearts to do. But we were mistaken. They even killed those who predicted the coming of the Righteous One." - Acts 7:51-52 The Jews have always been afraid of the radicals of God. They have always dwelled in the safety and understandings of the past and fear the effects of these radicals. But when these radicals like the prophets arrive, they persecute them because they are uncertain about the truth. An example is when the Jews didn't want to believe Stephen in Acts 7:56-58, " Look,' he said, I see heaven open and the Son of Man standing at the right hand of God.' At this they covered their ears and yelling at the top of their voices, they all rushed at him, dragged him out of the city and stoned him." A lot of potshots have been taken at the Church over the years. In spite of its obvious imperfections, the Church is the means that has been chosen to carry the message of the Gospel to the world. One wonders why a better system could not have been devised. Then one realizes that any system that has people in it is going to be imperfect. Alexander Solzhenitsyn wrote in the Gulag Archipelago that it was in prison where he learned that the line separating good and evil passes not through states, not through classes, not through political parties, either, but right through every human heart and through all human hearts. When we give ourselves serious evaluation, we find things hiding in our hearts that, if we could choose, we would remove. Our hearts have been described as "a zoo of lust, a bedlam of ambitions, a nursery of fears, a harem of fondled hatreds." Yet the Church, by its very nature, must be made up of the likes of us. "Imagine yourself as a living house," wrote C. S. Lewis. "God comes in to rebuild that house. At first, perhaps, you can understand what He is doing. He is getting the drains right and stopping the leaks in the roof and so on. But presently He starts knocking the house about in a way that hurts abominably and does not seem to make any sense. What on earth is He up to?

The Dangers of Isolation in The Catcher in the Rye It is normal to want to get away from all of the problems of the world, but it is not normal to want to be completely isolated from people. Holden wanted to have no human contact what so ever, and that is not normal. Throughout the book Holden expresses a rebellious attitude toward the world, and this rebellious attitude comes from his infatuation with being alone. He isolates himself from the world because he has not yet found himself and is searching. Holden feels that he must find himself alone with no one else's help. Holden expresses his rebellious side when he gets kicked out of school, again. Holden doesn't like school because he doesn't like doing activities that he loses patience for and sees no point in doing them. Holden also is rebellious in the way that he smokes and drinks when he is a minor. He is an excessive smoker and turns to alcohol to suppress his feelings of depression, which are signs of alcoholism. This behavior is not unheard of, but is rare for a 16-year-old to become an alcoholic. From this rebellious attitude Holden becomes isolated from those around him. His first act of isolation with a combination of rebellion is when Holden doesn't go to the game in the beginning of the book. Everyone was going to be there and he doesn't want to be like everyone else. Getting kicked out of school is another example of him rebelling and the cause of it being isolation. After he leaves Pency, Holden meets up with an old friend of his, Sally. After hanging out with her for awhile Holden asks her if she wants to run away with him. From this we learn that he has a desire to get away from the world. From this quote that Holden said we can tell how much he wants to run away, â€Å"I thought what I'd do was, I'd pretend I was one of those deaf-mutes. That way I wouldn't have to have any god dam stupid useless conversations with anybody. If anybody wanted to tell me something, they'd have to write it on a piece of paper and shove it over to me. They'd get bored as hell doing that after awhile, and then I'd be through with having conversations for the rest of my life†(p 198). Upon being kicked out of school, Holden decides to go on a little vacation. In this short period of time Holden goes through many tribulations. To get from place to place Holden takes taxicabs. During these rides Holden asks the cab drivers if they know where the ducks go when the pond freezes over. The reactions from the cab drivers are different each time, but his recurring concern about the ducks seems to be symbolic of Holden's desire for purpose and direction. While he is by himself, Holden doesn't stay in one place for very long. He didn't know where to go next just like he didn't know where his life was going. During his time by himself, Holden imagines becoming a deaf-mute and running away. Holden wants to use his imagination to feel more connected to the world and his emotions. He does this because in his fantasy world he can control what happens and in real life he wanted to do the same. Towards the end of the book, Holden loses more and more of his sense of reality. If he had stayed on this path he would have lost all sense of reality. All of these feelings that Holden had were each the cause of themselves. He kept going around in a destructive and unproductive cycle, which would be hard to break. In the end of the book, Holden didn't ride the merry-go-round because he wanted to break that cycle. He wanted to be there for his sister and see her grow up. He wanted to be a part of life, and the world. In order to do that he would need some help, and the mental hospital was the perfect place for the kind of help Holden needed.

The preceding argument is not entirely true as it is based on existing misconceptions about teenage outcasts. This is because not all teenage students who tend to exhibit behavioral isolation came from problematic families. In fact, in some instances such students came from good and well of families, and the isolation may have been caused by psychological factors such as shyness (Mosser, 2011). All that such students may be in need of is just some company so as to make them feel wanted and as part and parcel of the society. Stereotyping them thus only leads to long term negative impacts that would always place them at a disadvantage, one that they may wholly never recover from (U.S News Science, 2010). The second common stereotype revolves around the gay community. Majority of the people perceive gay people as having a tendency of exhibiting feminine traits. Any person who walks, talks, or engages in activities that are considered a preserve of the female gender are out rightly labeled as gay. It is not uncommon to hear of most people referring male fashion designers, stylists, or male hair dressers as gay. The st ereotype here is that all persons of the male gender who engage in professions that are considered feminine are gay. The stereotype here is even extended to ordinary male persons who exhibit feminine tendencies, or tend to lean more on female behavioral characteristics. This stereotype has been largely perpetuated by the media, which portrays all gay men as being extremely effeminate. So, are these stereotypes about gay men being effeminate true? Research suggests that gay men actually do prefer some professions like interior designing, fashion and hair coloring (Stossel and Binkley, 2006). In fact, professions like dancing and fashion require the creative touches of the gay people, and it is no wonder that such professions are flooded with gay men. Michael Bailey, a psychology professor, argues that these stereotypes of the gay men are actually true. He argues that gay men exhibited feminine tendencies during childhood and had a preference for feminine sex roles (Bailey, 2003). How ever, it is such stereotyping of the gay people that has led to homophobic attacks and discrimination among the gay community. Experiment on Changes in Serum Levels of Irisin Khaled Abdelfattah Abdelhamid Abulfadle, MD   Serum irisin level changes after propylthiouracil treatment of L-thyroxine induced hyperthyroidism in rats ABSTRACT Background: Thyroid hormones play an essential role in lipid, protein and carbohydrate metabolism; and they regulate thermogenesis and basal metabolic rate. Irisin is a myokine that is secreted as a product of fibronectin type III domain containing 5 (FNDC5) was shown to be present in many tissues, including the thyroid tissue. Objective: To investigate the possible changes in serum levels of irisin that occurred in a rat model of hyperthyroidism and the effects of use of propylthiouracil (PTU) in its treatment. Design: 18 male albino rats were divided into 3 groups. Group I was the intact control. Rats of group II and III received daily 0.3 mg/kg subcutaneous injection of L-thyroxine (LT4) for 12 consecutive days. On the thirteenth day, animals of group III (Hyperthyroid PTU treated) received 5 ml/kg i.p. injection of propylthiouracil (PTU) along with an equivalent amount of LT4 as administered to group II animals for more 15 days. Animals of group II (Hyperthyroid control) continued to receive LT4. On the day of termination (28th day of starting LT4 treatment), overnight (12hrs) fasted animals were sacrificed by decapitation under ether anesthesia. Results: In the hyperthyroid control group, there was a significant decrease in serum level of TSH (P0.05) in serum levels of TSH, T4, T3 irisin, and, BMI in hyperthyroid PTU group in comparison to that in the intact control group. In the hyperthyroid control group, there was a significant increase in the serum level of both glucose (P0.05) in the hyperthyroid PTU treated group in comparison to that in the intact control group. In the hyperthyroid control group, there was a significant decrease in serum levels of both TC (P0.05) in serum levels of both TG and LDL-C in comparison to that in the intact control group. Conclusion: This study showed a significant increase in serum levels of irisin in L-thyroxine induced hyperthyroidism in rats. On using PTU, serum levels of irisin, thyroid hormones, metabolic parameters and BMI were significantly changed in comparison to the hyperthyroid control group. Introduction: Irisin is a myokine that is secreted as a product of fibronectin type III domain containing 5 (FNDC5) in the skeletal muscle (Samy, Ismail et al. 2015, Jin Zhang 2017, Nathan C. Winn 2017). FNDC5, which is the precursor of irisin was shown to be present in many tissues, including the thyroid tissue (Huh, Panagiotou et al. 2012). It is secreted by a peroxisome proliferator-activated receptor ÃŽÂ ³ (PPARÃŽÂ ³) coactivator 1ÃŽÂ ± (PGC1-ÃŽÂ ±) after an exercise (Aydin 2014). Irisin was found to have an anti-obesity effect as it replaces the white fatty tissue with brown fatty tissue by the increase of uncoupling protein-1 (UCP1) resulting in oxygen consumption and stimulation of thermogenesis (Bostrà ¶m, Wu et al. 2012, Castillo-Quan 2012, Roca-Rivada, Castelao et al. 2013, Jin Zhang 2017, Nathan C. Winn 2017). Stengel, Hofmann et al. (2013) found that irisin level was identified to be high in obesity and to be low in anorexia nervosa. On the other hand, thyroid hormones pla y an essential role in lipid, protein and carbohydrate metabolism; and they regulate thermogenesis and basal metabolic rate (Hulbert 2000). Also, hyperfunction of thyroid gland is a common endocrine disorder which is accompanied by metabolic imbalance, oxidative stress and abnormal energy homeostasis (Erdamar, Demirci et al. 2008). There were some controversies in the results of studying the effect of thyroid hormone level changes on serum levels of irisin as Yalcin, Akturk et al. (2016) found an increase in serum irisin levels in hyperthyroidism and they suggested that it might contribute to altered energy metabolism. Also, Samy, Ismail et al. (2015) stated that hyperthyroidism was associated with up-regulation of serum irisin in male rats, probably as a response to myopathy and/or oxidative damage observed in both conditions. Moreover, Ruchala, Zybek et al. (2014) reported a borderline elevation of serum irisin in hyperthyroid versus hypothyroid patients. On the contrary, Panagiot ou, Pazaitou-Panayiotou et al. (2016) stated that changes in thyroid hormone levels did not affect circulating irisin levels in humans. Also, Stengel, Hofmann et al. (2013), Ellefsen, Vikmoen et al. (2014) and Gouni-Berthold, Berthold et al. (2013) reported lack of associations between levels of irisin, TSH and/or thyroid hormones. On the other hand, Zybek-Kocik, Sawicka-Gutaj et al. (2016) stated that irisin concentration was changed during prolonged thyroid function impairment. Also, AteÃ…Å ¸, Altay et al. (2016) found that, in hypothyroidism, there was a positive correlation between serum irisin levels and thyroid stimulating hormone (TSH) while a negative correlation was found between irisin levels and thyroxine (T4). Thus, this study was conducted to investigate the possible changes in serum levels of irisin that occurred in a rat model of hyperthyroidism and the effects of use of propylthiouracil (PTU) in its treatment. Materials and Methods: Animals preparations and experimental protocol: In the Department of Physiology, Faculty of Medicine, Zagazig University, this experimental study was done from 6th of November 2016 to 28th of December 2016. 18 adult male albino rats, age and weight matched (170-195 gm), were purchased from the animal house of Zagazig University and were housed at 20-22 °C on a 12-h light-dark cycle and for 5 days, to achieve acclimatization, they were supplied with tap water and a standard chow diet. Rats were divided into 3 groups of 6 each. Group I animals served as the intact control, whereas those of group II and III received daily subcutaneous injection of L-thyroxine (LT4) (Sigma, MO, USA) at a dose of 0.3 mg/kg for 12 consecutive days (Panda and Kar 2007). On the thirteenth day, animals of group III received propylthiouracil (PTU) (Sigma, MO, USA) 10 mg/kg intraperitoneally (Panda and Kar 2007) along with an equivalent amount of LT4 as administered to group II animals. PTU was intraperitoneally (i.p.) injected, in a volume of 5 ml/kg, dis solved in saline (Kim, Kim et al. 2012). Experiment was continued for more 15 consecutive days (from 13th LT4 treatment). Animals of group II continued to receive LT4 and acted as hyperthyroid control. Intact control rats were injected with 5 ml/kg/day subcutaneously with saline. On the day of termination (28th day of starting LT4 treatment), overnight (12hrs) fasted animals were sacrificed by decapitation under ether anesthesia. Blood from each animal was collected and by its centrifugation at 3000 rpm for 15 minutes, serum was separated and stored -20oc till the time of assay of different biochemical parameters (Kumar, Kar et al. 2014). PTU was used to treat hyperthyroidism by decreasing the amount of thyroid hormone produced by the thyroid gland (Nakamura, Noh et al. 2007), and it also inhibited the enzyme 5-deiodinase, which converts thyroxine (T4) to the active form triiodothyronine (T3) (Bahn, Burch et al. 2009). Also, it inhibited LT4-induced hyperthyroidisms in rats (Panda a nd Kar 2007). Body mass index (BMI) changes: BMI in gm/cm2 was calculated in the different groups, just before rats were sacrificed (under ether anesthesia), by dividing Body weight (gm) on Length2 (cm2) (Novelli, Diniz et al. 2007). A graduated (in centimeters) metal ruler was used for measuring the rat length by putting the zero end at the rat anus and recorded the reading which reached by the nose. BMI was used as a measure for the change in body weight of rats (Novelli, Diniz et al. 2007). Figure-1: Experimental design. Rats were divided into 3 groups. Group I was the intact control. Rats of group II and III received daily subcutaneous injection of levothyroxine (LT4) for 12 consecutive days. On the thirteenth day, animals of group III (Hyperthyroid PTU treated) received i.p. injection of propylthiouracil (PTU) along with an equivalent amount of LT4 as administered to group II animals for more 15 days. Animals of group II (Hyperthyroid control) continued to receive LT4. On the day of termination (28th day of starting LT4 treatment), overnight (12hrs) fasted animals were sacrificed by decapitation under ether anesthesia. Measurement of serum parameters: Serum level of irisin was estimated (in ng/ml) using an irisin rat ELISA assay kit following the manufacturers instructions (Catalog# EK-067-52; Phoenix Pharmaceuticals Inc., Burlingame, California, USA) with a detection range of 0.1-1000 ng/ml. Quantitative immunoassaying of T3, T4 and TSH using commercially available kits as recommended by the manufacturer (cat#SE120091, SE120090, SE120135, ELISA Kits, Sigma-Aldrich). Both serum levels of total cholesterol (TC) and triglycerides (TG) were estimated using enzymatic colorimetric methods. Serum high density lipoprotein-cholesterol (HDL-C) was assayed using NS Biotec HDL-precipitating reagent. Serum low density lipoprotein-cholesterol (LDL-C) was calculated using the Friedewald formula: LDL-C (mg/dl) = [(TC) (HDL-C) ( (Friedewald, Levy et al. 1972) Estimation of both serum glucose levels, using an automated analyzer (YSI 2300; YSI Life Sciences, Yellow Springs, OH), and serum insulin levels, using ELISA kits (RD Systems, Minneapolis, MN), were done. For assessing insulin resistance (IR), the homeostasis model assessment-IR (HOMA-IR) index was calculated by: HOMA-IR = (Nayak, Hillemane et al. 2014). There is a direct relation between insulin resistance and the value of HOMA-IR (Bonora, Targher et al. 2000). Statistical analysis: By the use of IBM SPSS Statistics for windows package version (24), data were analyzed and described as Mean ± SEM. For calculation of the statistical significance, One-way ANOVA and Tukey HSD post hoc test for multiple group comparison were used. P†°Ã‚ ¤0.05 indicated significance. Table-1 showed changes in serum levels of TSH, T4, T3 irisin, and, BMI among different groups. In the hyperthyroid control group, there was a significant decrease in serum level of TSH (0.81 ±0.05, P group (2.82 ±0.12), (0.54 ±0.01) (5.33 ±0.14), (1.29 ±0.04) and (401.67 ±5.65) respectively. On the other hand, there were insignificant changes (P>0.05) in serum levels of TSH, T4, T3 irisin, and, BMI in hyperthyroid PTU group in comparison to that in the intact control group. Also, there was a significant increase in both serum level of TSH (2.68 ±0.14, P0.05) in the hyperthyroid PTU treated group in comparison to that in the intact control group. Moreover, there was a significant decrease in the serum levels of both glucose (91.33 ±1.2, P0.05) in serum levels of both TG (52.33 ±0.67) and LDL-C (36.03 ±3.07) in comparison to that in the intact control group (106.5 ±2.51), (54 ±1.15), (55.17 ±1.22) and (41.47 ±3.27) respectively. In the hyperthyroid PTU treated group, there was a significant increase in serum levels of both TC (103.33 ±1.63, P0.05) in serum levels of both TG (53.17 ±0.95) and LDL-C (41.37 ±1.92) in comparison to that in the intact co ntrol group (89.83 ±2.52), (43.33 ±0.88), (52.33 ±0.67) and (36.03 ±3.07) respectively. Also, there were no significant changes (P>0.05) in serum levels of TC, TG, HDL-C and LDL-C in the hyperthyroid PTU treated group in comparison to that in the intact control group. Figure-2A showed a significant positive correlation between serum irisin levels and that of T4 (r=0.97, P Discussion: This study was done to declare changes in irisin serum levels in a rat model of hyperthyroidism and the effect of PTU therapy on it. Also, to identify the association between serum irisin level changes in such conditions and some metabolic parameters including insulin resistance and lipid profile. The results of this study showed a significant decrease in serum level of TSH and BMI, but, a significant increase in serum levels of T4, T3, and irisin in the hyperthyroid control group, in comparison to that in the intact control group. Also, there was a significant positive correlation between serum irisin levels and that of T4 T3 but a significant negative correlation with serum TSH in the hyperthyroid control group. These results were supported by Huh, Panagiotou et al. (2012) who stated that FNDC5, irisin precursor, was present in the thyroid tissue. Also, the present study results were supported by Irrcher, Adhihetty et al. (2003), Ruchala, Zybek et al. (2014), Sanchis-Gomar and Per ez-Quilis (2014), Samy, Ismail et al. (2015) and Yalcin, Akturk et al. (2016) who found an increase in serum irisin levels in hyperthyroidism and they suggested that it might contribute to altered energy metabolism. Moreover, Irrcher, Adhihetty et al. (2003) reported that T3 increased the irisin precursor, PGC-1ÃŽÂ ±, expression in muscle. Furthermore, Sanchis-Gomar and Perez-Quilis (2014) stated that serum irisin was increased in hyperthyroidism as a compensatory response to oxidative stress as it had an antioxidant effect. On the contrary, Gouni-Berthold, Berthold et al. (2013), Stengel, Hofmann et al. (2013) and Panagiotou, Pazaitou-Panayiotou et al. (2016) stated that changes in levels of thyroid hormones did not affect circulating irisin levels in humans. This discrepancy between their results and that of this study could be explained by species difference. Regarding the negative correlation between both irisin and TSH hormones which was present in this study, it was support ed by Ruchala, Zybek et al. (2014). On the contrary, Stengel, Hofmann et al. (2013) and Ellefsen, Vikmoen et al. (2014) reported lack of correlation between serum levels of irisin and that of TSH and thyroid hormones. Regarding body weight changes with hyperthyroidism, the result of this study was supported by Janson, Karlsson et al. (1995), Voldstedlund, Tranum-Jensen et al. (1995) and Loeb (1996) who found a decrease in body weight with hyperthyroidism. Also, this study results were supported by Bostrà ¶m, Wu et al. (2012), Castillo-Quan (2012) and Roca-Rivada, Castelao et al. (2013) who found that irisin had an anti-obesity effect as it replaced the white adipose tissue with brown adipose tissue and increased oxygen consumption and thermogenesis. On the contrary, Stengel, Hofmann et al. (2013) and Saleh, et al. (2014) found that irisin level was high in obesity and low in anorexia nervosa. This discrepancy between their results and that of this study can be explained by species difference. On the other hand, this study results showed insignificant changes in serum levels of TSH, T4, T3 irisin, and, BMI in hyperthyroid PTU group in comparison to that in the intact control group. This indicated the improvement that occurred with PTU therapy in cases of hyperthyroidism which was supported by Nakamura, Noh et al. (2007), Panda and Kar (2007) and Bahn, Burch et al. (2009). Also, the results of this study showed a significant increase in both serum level of TSH and BMI, but, a significant decrease in serum levels of T4, T3, and irisin in the hyperthyroid PTU treated group in comparison to that in the hyperthyroid control group. These results were supported by Park, Lee et al. (2016) who found that PTU administration ameliorated hyperthyroidism, reducing T4 and T3, and, increasing both TSH and BMI. Moreover, the results of this study showed a significant increase in the serum level of both glucose and insulin, and, the value of HOMA-IR in the hyperthyroid contro l group in comparison to that of the intact control group. Also, there was a significant positive correlation between serum irisin levels and that of glucose in the hyperthyroid control group. These results indicated occurrence of insulin resistance with hyperthyroidism which was supported by Loeb (1996). This was supported also by Saleh, et al. (2014) who stated that hyperglycemia gradually up regulated FNDC5/irisin expression in the skeletal muscles non-diabetic persons. Also, this study results were supported by Hee Park, Zaichenko et al. (2013), Liu, Wong et al. (2013) and Mehrabian, Taheri et al. (2016) who found a positive correlation between serum irisin and that of glucose. On the contrary, Mehrabian, Taheri et al. (2016) found that serum irisin was negatively correlated with serum glucose level. Opposite to the results of this study, Huh, Panagiotou et al. (2012), Choi, Kim et al. (2013) and Sanchis-Gomar and Perez-Quilis (2014) found that decreased blood irisin level was a ccompanied by insulin resistance development. Sanchis-Gomar and Perez-Quilis (2014) stated also, that irisin ameliorated insulin resistance by increasing betatrophin hormone expression which stimulated ÃŽÂ ²-cell regeneration. Furthermore, the results of this study showed no significant changes in the serum level of both glucose and insulin, and, the value of HOMA-IR in the hyperthyroid PTU treated group in comparison to that in the intact control group. Also, there was a significant decrease in the serum levels of both glucose and insulin, and, the value of HOMA-IR in the hyperthyroid PTU treated group in comparison to that of the hyperthyroid control group which means improvement of insulin resistance with PTU treatment. On the other hand, Huh, Panagiotou et al. (2012) found that exposure to high blood glucose for a long time was associated with a significant decrease in irisin serum level. The discrepancy between their results and the result of this study could be explained by the difference in species and duration of the study. In the hyperthyroid control group, there was a significant decrease in serum levels of both TC and HDL-C but, there were insignificant changes in serum levels of both TG and LDL-C in comparison to that in the intact control group. Also, there were significant negative correlations between serum irisin levels and that of both TC and HDL-C in the hyperthyroid control group. On the other hand, in the hyperthyroid PTU treated group, there was a significant increase in serum levels of both TC and HDL-C but, there were insignificant changes in serum levels of both TG and LDL-C in comparison to that in the intact control group. Also, there were no significant changes in serum levels of TC, TG, HDL-C and LDL-C in the hyperthyroid PTU treated group in comparison to that in the intact control group. The results of this study was partly supported by Mehrabian, Taheri et al. (2016) found a negative correlation between irisin and TC but, a po sitive correlation with HDL-C in normal weight obesity. On the other hand, Liu, Wong et al. (2013) found a positive correlation between serum irisin level and TC in non- obese, non- diabetic persons. Also, Sanchis-Gomar, Alis et al. (2014) reported insignificant correlation between irisin and TC. Conclusion   Ã‚   This study showed a significant increase in serum levels of irisin in L-thyroxine induced hyperthyroidism in rats and this could be related to changes in thyroid hormones, body weight and metabolic parameters. On using PTU, serum levels of irisin, thyroid hormones, metabolic parameters and BMI were significantly changed in comparison to the hyperthyroid control group and insignificantly changed in comparison to the intact control group which confirmed the improvement of the hyperthyroidism. Future studies are needed to confirm these results and to ensure the possible use of irisin as a biomarker for proper treatment of hyperthyroidism.
